传播木尔坦棉花曲叶病毒的烟粉虱隐种鉴定

中文摘要:为明确广东地区传播木尔坦棉花曲叶病毒(Cotton leaf curl Multan virus,CLCuMuV)的烟粉虱隐种,采用分子鉴定、烟粉虱传毒试验与分子检测的方法,对传播CLCuMuV的烟粉虱隐种进行了鉴定.结果表明:在广东棉花、红麻和黄秋葵曲叶病发生的田间,2014年采集的30头烟粉虱mtCOI与Asia II 7隐种序列同源性为98.3%~99.6%,2015年采集的10头烟粉虱中,8头烟粉虱mtCOI与MEAM1隐种序列同源性为99.2%~99.5%,2头烟粉虱mtCOI与Asia II 7隐种序列同源性为99.8%~99.9%,说明烟粉虱种群包括入侵隐种MEAM1和土著隐种Asia II 7.在利用烟粉虱人工传毒试验中,MEAM1、Asia II 7和Asia II 1这3个隐种均可在棉花曲叶病株上饲毒获得CLCuMuV及β卫星分子;除MEAM1隐种外,Asia II 7、Asia II 1隐种可传播CLCuMuV,侵染红麻及黄秋葵植株引起曲叶病,前者传毒效率分别为50%和100%;而后者为33%和100%.以上3个烟粉虱隐种传毒接种棉花未见成功.

Abstract:In order to clarify the cryptic species of the whitefly Bemisia tabaci transmitting Cotton leaf curl Multan virus (CLCuMuV) in Guangdong Province, molecular identification, whitefly transmission and molecular detection were conducted in the study. The results indicated that, in the fields occurring leaf curl disease of upland cotton Gossypium hirsutum, okra Abelmoschus esculentus and kenaf Hibiscus cannabinus, mtCOI sequences of all 30 individual whitefly adults collected from the diseased plants in the field in 2014 showed the nucleotide sequence identities of 98.3%-99.6% with that of Asia II 7; among ten individual adults collected in 2015, mtCOI sequences of eight whitefly individual adults had the nucleotide sequence identities of 99.2%-99.5% with that of MEAM1, and the sequences of two whitefly adults had 99.8%-99.9% identities with that of Asia II 7. The consequence implied that the whitefly field populations consisted of two cryptic species, invasive MEAM1 and indigenous Asia II 7, in Guangdong Province. The whitefly transmission experiment showed that the three cryptic species MEAM1, Asia II 7 and Asia II 1 were able to acquire CLCuMuV and beta-satellite from the diseased cotton plants infected by CLCuMuV, and with the exception of MEAM1, Asia II 7 and Asia II 1 cryptic species were able to transmit CLCuMuV to infect kenaf and okra plants. The transmission efficiency of the former was 50% and 100%, while that of the latter was 33% and 100%, respectively. No infected CLCuMuV cotton plant transmitted by three cryptic species was found.
