中文摘要:为明确普通大蓟马Megalurothrips usitatus的光谱敏感性以及视蛋白基因潜在的行为调控机制,通过扫描电镜观察普通大蓟马雌、雄成虫复眼的外部形态特征,利用自制的行为测定装置测定该害虫在昼夜节律、单一光源和多种光源下的趋光性,采用实时荧光定量PCR技术分析4个视蛋白基因在普通大蓟马成虫不同组织及UV-A不同处理时间后的表达量变化。结果表明:普通大蓟马雌、雄成虫复眼外部形态特征无明显差异,雌、雄成虫单侧复眼大约各有70个小眼。普通大蓟马在白天(08:00—17:00)对UV-A、蓝光、绿光和白光4种单色光源的趋光率差异显著,而夜间(21:00—05:00)则无明显差异。普通大蓟马对白光和UV-A的趋光率在白天与夜间差异显著,其对蓝光和绿光的趋光率在白天与夜间大部分时间无显著差异。在4种单色光源组合中,普通大蓟马对UV-A的趋光率最大。4个视蛋白基因均在普通大蓟马成虫头部高表达,UV-A照射能显著诱导4个视蛋白基因上调表达。表明复眼类型影响普通大蓟马的趋光行为,且趋光行为主要受UV-A波长影响,同时UV-A还可以诱导视蛋白基因的上调表达。 |

Abstract:To clarify the spectral sensitivity of Asian bean thrips Megalurothrips usitatus and the potential behavior regulation mechanism of opsin genes, the external morphological characteristics of the compound eyes of female and male adults of M. usitatus were observed with scanning electron micro-scope. Phototaxis rates were measured using a self-made behavior measuring device under circadian rhythms, single light sources and multiple light sources. The expression changes of four opsin genes in different tissues of M. usitatus adults and under different UV-A treatment durations were analyzed by real-time fluorescence quantitative PCR. The results showed that there was no significant difference in the external morphological characteristics of compound eyes between female and male adults of M. usitatus, and there were about 70 ommatidia in each unilateral compound eye. During the daytime (08: 00—17: 00), phototaxis rates of M. usitatus differed significantly to across four monochromatic light sources, whereas no significant difference was observed at night (21: 00—05: 00). Significant differences in phototaxis rates between white light and UV-A were noted during day and night, while responses to blue and green light showed no significant differences most of the time. Among different combinations of monochromatic light sources, M. usitatus exhibited the strongest attraction to UV-A. The four opsin genes were highly expressed in the head of M. usitatus adults, and UV-A irradiation significantly induced the up-regulation of these genes. These findings suggest that the compound eye type of M. usitatus affects phototaxis behavior, with UV-A wavelength playing a dominant role, and UV-A also induces the up-regulation of opsin gene expression. |
