中文摘要:为提高植物源药剂对梨小食心虫Grapholita molesta的防治效果,测定饲喂含抑肽酶的人工饲料后梨小食心虫2龄幼虫中肠内总蛋白酶、胰蛋白酶和胰凝乳蛋白酶的活性,采用饲料混毒法测定0.20 mg/L抑肽酶对1%印楝素水分散粒剂和0.3%苦参碱水剂的增效作用。结果表明:饲喂含0.20 mg/L抑肽酶的人工饲料12 h后,梨小食心虫2龄幼虫中肠内总蛋白酶、胰蛋白酶和胰凝乳蛋白酶的活性均与对照无显著差异;饲喂36 h后,这3种蛋白酶的活性均较对照显著下降。当0.20 mg/L抑肽酶分别与LC10、LC30、LC50的1%印楝素水分散粒剂混配饲喂后,梨小食心虫2龄幼虫的实际死亡率分别为29.16%、43.06%和66.67%,均显著高于预测死亡率;当0.20 mg/L抑肽酶分别与LC10、LC30和LC50的0.3%苦参碱水剂混配饲喂后,梨小食心虫2龄幼虫的实际死亡率分别为44.44%、58.33%和68.89%,均显著高于预测死亡率。表明抑肽酶通过抑制梨小食心虫中肠蛋白酶活性对印楝素或苦参碱产生增效作用。 |

Abstract:To improve the control effect of botanical pesticides on oriental fruit moth Grapholita molesta, the activities of total protease, trypsin and chymotrypsin in the midgut of the second larvae of G. molesta were measured after feeding an artificial diet containing 0.20 mg/L aprotinin. The synergistic effect of 0.20 mg/L aprotinin on 1% azadirachtin WG and 0.3% AS was determined by feeding on the diet containing these toxic substances. The results showed that after feeding the artificial diet containing aprotinin for 12 h, the activities of total protease, trypsin and chymotrypsin in the midgut of the second instar larvae were not significantly different from the control group. After feeding for 36 h, the activities of these three proteases decreased significantly compared with the control group. When 0.20 mg/L aprotinin was mixed with LC10, LC30 and LC50 of 1% azadirachtin WG, the actual mortality rates were 29.16%, 43.06% and 66.67%, respectively, which were significantly higher than the predicted mortality. When 0.20 mg/L aprotinin was mixed with LC10, LC30 and LC50 of 0.3% matrine AS, the actual mortality rates were 44.44%, 58.33% and 68.89%, respectively, which were significantly higher than the predicted mortality. These results indicate that aprotinin has a synergistic effect on azadirachtin or matrine by inhibiting the activity of the midgut proteases in G. molesta. |
