中文摘要:为明确醛脱氢酶(aldehyde dehydrogenase,ALDH)在玉米病原真菌拟轮枝镰孢Fusarium verticillioides生长发育及致病中的作用,利用实时荧光定量PCR(quantitative real-time polymerase chain reaction,qRT-PCR)技术分析拟轮枝镰孢的ALDH家族成员FvALDH-52和FvALDH-82基因在侵染玉米籽粒不同时期的表达水平,通过遗传转化方法对FvALDH-52和FvALDH-82进行基因敲除,分析2个基因在拟轮枝镰孢生长发育及致病性中的作用。结果表明:FvALDH-52和FvALDH-82在侵染玉米籽粒过程中表达水平均升高。敲除FvALDH-52和FvALDH-82基因后,拟轮枝镰孢的分生孢子产生能力降低,同时对H2O2和乙醛更加敏感,产生的伏马毒素显著降低;FvALDH-52基因敲除后导致病菌对玉米籽粒和玉米茎秆的致病力减弱。表明ALDH基因FvALDH-52在拟轮枝镰孢生长发育和致病过程中起着重要作用。 |

Abstract:To clarify the function of aldehyde dehydrogenase (ALDH) in the growth, development, and pathogenicity of the maize pathogenic fungus Fusarium verticillioides, quantitative real-time polymerase chain reaction (qRT-PCR) was employed to analyze the expression levels of ALDH family members, the FvALDH-52 and FvALDH-82 genes of F. verticillioides, during different phases of its infection. Genetic transformation was carried out to knock out the FvALDH-52 and FvALDH-82 genes, with the aim of exploring the functions of these two genes in the growth, development, and pathogenicity of F. verticillioides. The results demonstrated that the expression levels of FvALDH-52 and FvALDH-82 increased during the infection process of maize kernels. Following the knockout of FvALDH-52 and FvALDH-82, the sporulation capacity of F. verticillioides decreased, and it became more sensitive to H2O2 and acetaldehyde, while the concentration of fumonitoxin declined substantially. The knockout of of FvALDH-52 weakened the pathogenicity of the pathogen towards kernels and stalks. These findings indicate that FvALDH-52 plays a crucial role in the growth, development, and pathogenicity of F. verticillioides. |
